The Thymidine Pool in Grasshopper Neuroblasts during Mitosis

چکیده

In an earlier study (10), it was shown that exogenous thymidine is incorporated into a precursor "pool" during the period between deoxyribonucleic acid (DNA) syntheses. The possibility was not examined, however, that the ability of cells to incorporate thymidine into the pool varied with the mitotic stage at the time of exposure. Available evidence indicates that the ability of cells to incorporate thymidine into a precursor pool may vary with cell stage. First, in situ synthesis of

Distribution of Tritium-labeled Dna among Chromosomes during Meiosis

Thymidine-H(3) of high specific activity was used to study the distribution of labeled chromatids during meiotic divisions in spermatocytes of a species of grasshopper (Orthoptera). The distribution is regularly semiconservative as has been shown previously for mitosis, i.e., all chromatids are labeled after incorporation of thymidine-H(3) into DNA at premeiotic interphase. If incorporation occ...

Timing of identity: spatiotemporal regulation of hunchback in neuroblast lineages of Drosophila by Seven-up and Prospero.

Neural stem cells often generate different cell types in a fixed birth order as a result of temporal specification of the progenitors. In Drosophila, the first temporal identity of most neural stem cells (neuroblasts) in the embryonic ventral nerve cord is specified by the transient expression of the transcription factor Hunchback. When reaching the next temporal identity, this expression is sw...

The Effects of Varying Concentrations of Colchicine on the Progression of Grasshopper Neuroblasts into Metaphase

The effects of four concentrations of colchicine (2.5 x 10(-7), x 10(-5), x 10(-3), and x 10(-2)M) on the cell cycle of grasshopper neuroblasts have been determined by direct observations on living cells. The lowest concentration, 2.5 x 10(-7)M, does not completely disorganize the spindle but does retard its action. The three higher concentrations disorganize the spindle, so that all cells reac...

Mitosis in primary cultures of Drosophila melanogaster larval neuroblasts.

Although Drosophila larval neuroblasts are routinely used to define mutations affecting mitosis, the dynamics of karyokinesis in this system remain to be described. Here we outline a simple method for the short-term culturing of neuroblasts, from Drosophila third instar larvae, that allows mitosis to be followed by high-resolution multi-mode light microscopy. At 24 degrees C, spindle formation ...

Extrinsic cues, intrinsic cues and microfilaments regulate asymmetric protein localization in Drosophila neuroblasts

BACKGROUND The Drosophila central nervous system develops from stem cell like precursors called neuroblasts, which divide unequally to bud off a series of smaller daughter cells called ganglion mother cells. Neuroblasts show cell-cycle-specific asymmetric localization of both RNA and proteins: at late interphase, prospero RNA and Inscuteable, Prospero and Staufen proteins are all apically local...
